Introduction
I wish I had someone to pretty this guide up for me, because I know what to say but not how to catch someone's eyes Q.Q
Flat Ability Power works best with Vel'Koz. This lets him be a threat pre 6 even when you spend you first 1,000 gold or so on a Chalice of Harmony and some boots.
I chose Armor Runes over Magic Resist because I believe Vel'Koz can beat any AP champ that doesn't really on their Auto Attack early game for damage. This, and the mandatory Chalice of Harmony that already gives Magic Resist.

Flash and Barrier are both musts. There is no alternative situation where maybe Ghost would be better, or maybe ignite over Barrier. I say this because of how Vel'Koz works:
Flash is better than Ghost BECAUSE of his low speed. It's like being overweight. It takes too much time and effort to try and fix his slow speed, rather than to embrace it. Flash provides Vel'Koz with the getaway from the ganks you will be getting. Vel'Koz is not the chasing type of champion, when enemies run away out of range of you Ult, simply let them. Think of Flash as the difference from starting 1/0/0, and 1/1/0.
Barrier is a must over Ignite for two simple reasons. One; if you are close enough to an enemy champion to use ignite, you are playing Vel'Koz wrong. Two; Vel'Koz is a BURST champion, ignite takes too long to do damage, where barrier provides the extra second or two for your next skill to come off cooldown and secure the kill!

Vel'Koz has one of the simplest, and strongest combos. When you see an enemy champion walking towards somewhere very predicably, cast your E and follow with your W immediately. If done right, your E and W should hit at the same time, and the knock-up from E will give your W enough time to hit the enemy champion twice! You NEED to cast your W immediately after E or it will not have the time to hit twice. If you miss your spells, run and say oh well. This simple two spell combo can easily bring any non-tank to half health because of Vel'Koz' passive, which deals true damage when you land three spells.
Now when you want to completely go ham on a champion and burst them from FULL health to ZERO health, do this! Cast your E W combo, and immediately follow up with Q then R. When done right, your Q should hit the enemy champion whilst they are still in the air, and your R should be hammering them when they hit the ground. At this point, most enemies will panic at the crazy amount of damage you delivered in all of about one second and they will either try to run, or try to attack back. If they try to run, follow them with your mouse and laugh while they die. If they try to attack you, you need to pay attention to if they get close and out of your laser. If the champion gets behind you or to your side, cancel your ultimate by pressing R again, then click away from them to lure them towards one direction and cast your W which will be available and your should have the kill. For the especially tanky champs, you may need to run a little more for your Q to come off cool down so you can for sure seal the kill.
