Life steal does not work on on-hit effects as it it's self is basically an on-hit effect.

As others above me have mentioned Life steal doesn't work for abilities which do not proc on-hit effects - this includes proc items like Wits End and Madred's.

So if I understand you well, Life Steal works only with flat physical damage. In this case, to increase the amount of HP drain with Life Steal I must seek to increase my base attack damage and/or my Attack speed. Is that correct?

Just to be 100% clear, it doesn't apply on abilities that deal physical damage, only on autoattacks.
Except for a few special abilites like
Parrrley and Mystic Shot.
Auto attack physical damage = life steal.
Crit damage will work for life steal (I bring it up because the word flat was used.)
All non "On hit" or "Apply to next hit" abilities = spell vamp.
GP Q = Life steal.
Nasus Q = Life steal.
Viegar's skills = Spell Vamp.
Annie's spells = Spell Vamp.
Etc.
